Five hundred years ago, a feudal lord was attacked by demons wanting to retrieve his mysterious inner power, the invincible power that can help them take over the human world. The famous mage, Tokimori Hazama, was called upon, and with his 2 students, managed to save the whole land. But they fell ill, the feudal lord was killed, his power was also buried with him at a site called Karasumori.
In the present day, Yoshimori Sumimura and Tokine Yukimura, heirs of the Hazama clan, became the kekkaishi protecting the land and the power. They use a technique called Kekkai, a form of magical barrier, used to capture and destroy Ayakashi (monsters/demons) that are drawn to this land. Any demons that come near the power will become stronger and therefore Yoshimori and Tokine are to guard the land from intrusion of the Ayakashi.
But some of the demons form a powerful organization to take over that mysterious power. Yoshimori plans to destroy not only the demons, but also to gain the power to completely save the world from being controlled by demons. To do that, he first needs to know what the power actually is, and also strengthen his poor kekkai skills.

In the film, Yūko receives a request from a woman to help her back into her mansion which for some reason does not allow her to enter. Yūko says since Watanuki was the one who brought the client to her, Watanuki should be the one to grant her wish, and therefore brought along. Dōmeki is also brought along to help and Yūko attends not only to grant her client's wish but also because she is invited to the mansion where many famous collectors were also invited with the same letter. Inside the mansion are strange rooms and mysterious letters telling its guests what do to in place of a host. Though the collectors think up the theory of a mysterious auction being held like in an urban legend, it becomes apparent that something more sinister and mysterious going on. One after another, each collector disappears and a number of other strange occurrences happen during the night. The real reasons behind the invitations must be discovered in order for Yūko and company to get to the bottom of this mystery.

Info (from Wiki): One day while heading home, Ageha Yoshina, a high-schooler who helps people with their problems for 10,000 yen, finds a public pay phone ringing by itself. He picks it up to get no answer except a calling card with the word "Psyren" printed on it. After Sakurako Amamiya, a girl in Yoshina's class, disappears after mention "Psyren", Yoshina uses the calling card in hopes of finding the whereabouts of his classmate, despite being warned not to get involved. The next day, while running from men sent to collect the calling card, Yoshina is sent to another world known as Psyren. Once in this new world, Yoshina finds Amamiya and others that have been sent to this odd place. When most of the group is annihilated by a masked monster, Yoshina figures that he has to work with his acquaintances in order to make it back home.

Kimihiro Watanuki is a high school student plagued by yōkai, supernatural beings magically attracted to him. The creatures are invisible to others, and as a result he feels extremely burdened by them. When he quite literally stumbles into a shop that grants wishes, his life changes drastically.
The shop is owned by Yūko Ichihara, a mysterious, beautiful witch of many names and esoteric renown. The seemingly all-knowing witch offers to grant Watanuki's wish to be rid of his ability to see spirits. However, in exchange, he must pay a price equal to the power required to grant the wish. Yūko discovers that Watanuki is very skilled with his hands and a hard worker. As a result, Watanuki is forced to perform housework and odd jobs for Yūko until he has done enough work for his wish to be granted.
Shortly afterwards, other characters become involved with Watanuki's position, including his love interest, Himawari Kunogi, and his "rival," Shizuka Dōmeki. The three become increasingly "friendly," despite the number of times Watanuki becomes annoyed by Dōmeki's calm demeanor.
